Leymus chinensis,a high quality grazing,adapted to alkaline-sodic soil conditions and distributes widely in the Northeast of China.The species have developed molecular and physiological mechanisms to adapt itself under such stress conditions during the course of evolution.The mechanisms must involve lots of anti-stress genes and proteins.The full length of the LcFERgene was cloned by rapid-amplification of cDNA ends(RACE).The sequence was analysed by tools on NCBI.pGEX-KG-LcFER plasmid was obtained after digestion using BamH I and XbaI restriction endonuclease sites.Recombinant fusion protein was produced after transformation into E.coli BL21(DE3)with IPTG induction.The results showed that the coding sequence of LcFERwas 729bp.The amino acid sequence of LcFERshowed high identity with FER gene of Triticm aestivum,Hordeum vulgare,Oryza sativa.The LcFERcontained three consensus function domain,the ferroxidase diiron center,ferrihydrite nucleation center and iron ion channel.The secondary structure analysis revealed that LcFERstrongly resembled other FERgenes.SDS-PAGE identification was also used to analyse the recombinant protein expression,and a fussion protein band visualized successfully.The results establish a basis for further functional research of LcFERgene.
