Objective:Ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco)from Chrysanthemum morifolium was cloned in silico cloning.rbcLgene sequences in silico cloning,properties of encoding amino acid and adaptive evolution were analyzed.Method:According to the related literature,a full-length rbcLgene sequences form Chrysanthemum morifolium was cloned in silico cloning,and the structure and function of rbcLprotein were predicted by bioinformatics analysis.In the site-specific model,we found amino acid positive selection sites inrbcLgene.Result:A complete open reading frame sequences of electronic cloning chrysanthemumrbcLgene was 1 452 bp and encoded 483 amino acids.rbcLprotein secondary structure has 19α-helices and 18β-sheets.Seven amino acid sites(228S,255 V,279T,309 M,328S,439 T,449T)were found under positive selection.Conlusion:The complete sequence fromChrysanthemum morifolium rbcLgene in silico cloning provided certain reference data for further study,and encoded protein adaptability research was helpful to explore the mechanism of compositae adapting to the environment.
