Sequencing of Whole Genome of Human Parainfluenza Virus Type 3 Strain LZ22 Isolated in Lanzhou City,China

Abstract
Objective To sequence the whole genome of human parainfluenza virus type 3(HPIV-3)strain LZ22 isolated in Lanzhou City,China.Methods The nucleotide of whole genome of LZ22 strain was sequenced by genomic walking method and RACE,based on which the structural character of and evolution of the strain were evaluated.Results The whole genome of LZ22 strain consisted of 15 462 nucleotides,which complied withrule of six.The nucleotides encoded 6 structural proteins in an order of 3'-NP-PP/C-M-F-HN-L-5'.The genomic structure of LZ22 strain was identical to that of HPIV-3 strain with known sequence.However,the genomic comparison with other 4 HPIV-3 reference strains in GenBank revealed the highest homology of 99%(with 147 nucleotide variation)of LZ22 strain to ZHYMgz01 strain isolated in Guangzhou City,China and the lowest homology of 94.6%(with 832 nucleotide variation)of the strain to 14702 strain isolated in Canada.Phylogenetic analysis showed the closest evolutionary relationship of LZ22 strain with ZHYMgz01 strain,second with GP strain(Japan),and the farthest relationship with 14702 and JS(USA) strains.Conclusion LZ22 strain was of the genetic character of paramyxovirus,of which the nucleotide sequence was highly homologous to that of HPIV-3 strain.The phylogenesis of HPIV-3 strain may be correlated with geographical region.
