Study on the Interaction Between Luteolin and Bovine Serum Albumin by Fluorescence Spectroscopy

Abstract
The interaction behavior between luteolin and bovine serum albumin(BSA) in physiological buffer (pH=7.4) was studied by fluorescence spectroscopy. The quenching constants were obtained at 292 K,299 K and 311 K. The results indicated that the fluorescence quenching mechanism for BSA through luteolin binding was a static quenching process. The thermodynamic parameters,enthalpy change (△H) and entropy change (△S),were calculated to be-4.755 kJ\5moL-1 and 93.2 J\5moL-1\5K-1 respectively,which indicated that the interaction of luteolin with BSA was driven mainly by electrostatic force. The process of binding was a spontaneous process in which Gibbs free energy change was negative. Further more,the effect of luteolin on the conformation of BSA was investigated using synchronous fluorescence spectroscopy.
