In vitro fertilization and intracytoplasmic sperm injection using frozen boar spermatozoa

Abstract
Frozen boar spermatozoa were used to conduct in vitro fertilization(IVF) and intracytoplasmic sperm injection(ICSI) in swine.The effect of addition of L-cysteine into medium and different culture method for maturation was also investigated.The results showed that ICSI resulted in significantly lower cleavage rate and blastocyst percentage(68.3%versus 76.9%and 11.4% versus 19.2%,respectively(P0.05) than those from IVF group.Addition of 1.71 mmol·L~(-1) L-cysteine into medium to culture ICSI embryos for 3 h improved neither cleavage rate as well as developmental rate to morula nor blstocyst percentage significantly (P0.05).Using 0.4%BSA NCSU-23 medium for the maturation of porcine oocytes seemed to yield higher cleavage rate of ICSI embryos than other groups.However,the developmental rate of embryos from each group had no differences(P0.05),indicating that the maturation method might not impact the development of ICSI embryos.After co-culture with granulosa cells or cumulus cells,the cleavage rate of ICSI embryos was improved significantly than that of the control(P0.05),but there is no statistic difference in the blastocyst percentage among groups.
