STUDY ON THE EFFECTS OF COMBINED NUTRIENTS ON COGNITION AND MOTOR FUNCTION AND ITS MECHANISM IN AGED RATS

Abstract
Objective: To investigate the improved effects of combined nutrients in aged rats with impaired cognition and motor function and illustrate its possible mechanism. Method: Twenty four Wistar rats were randomly divided into intervened group(15 mo old) and control group(3 mo old), six male and six female in each. The rats of intervened group were fed with diets supplemented with combined nutrients while the control group with basic diet. 4ml/ (kg FF*bw FF*d) mixed plants extract and water were administered intragastically to intervened and control group respectively. The experiment lasted 10 w. The cognition and motor behavior were tested with water maze and rod walking test. The level of acetylcholine in brain was determined by alkaline hydroxylamine colorimetry and noradrenaline, serotonin and dopamine by fluorospectrophotometry. The activity of acetylcholine esterase in serum and brain was tested according to the instruction of kits. The changes of long-term potentiation of hippocampal dentate gyrus were observed through electrophysiology. Results: Compared with control group, the cognition and motor function of aged rats in intervened group were improved. The contents of acetylcholine, noradrenaline, serotonin and dopamine in brain were increased while acetylcholine esterase activity lowered in serum and brain. Theamplitude of LTP of hippocampal dentate gyrus and the efficacy of hippocampal synaptic transmission were significantly elevated. Conclusion: The combined nutrients have improved effects on cognition and motor function in aged rats, and the mechanism may be correlated with elevated neurotransmitter level and efficacy of synaptic transmission.
