Abstract
The extracting condition and electrophoresis method of millet alcohol-soluble protein was studied based on wheat gliadin gel electrophoresis method.The results indicated that the extraction of prolamin was most efficient with extraction solution composed of isopropyl alcohol(50%) and β-mercaptothanol(1%) at the temperature of 60 ℃ for 30 minutes.The clear and sepereated alcohol-soluble protein bands were obtained with the concentration of acrylamide at 12.5% and the electrode solution composed of 0.4‰ glycin and 8‰ acetic acid glacial.It was a efficient electrophoresis method to identify and evaluate millet varieties.
