Abstract
The factors influencing on the aseptic culture of Phalaenopsis hybrid seeds were studied.The results showed that sowing method,culture medium,light time,temperature and cross combination had obviously effects on the yield of protocorm body (PB).Sowing method,culture medium and temperature had significant effects on 100-grain weight of PB.Organic additives had significant effects on the growth and development of PB.The suitable culture method for the hybrid seeds of different ploidy Phalaenopsis was as follow:seeds diluted with aseptic water were sown on 1/2 MS medium supplemented with 6-BA 0.2 mg/L,NAA 0.2 mg/L and CM 100 ml/L and cultured at 27 ℃ under light intensity of 20.27-13.51 μmol/(m2·s) and five hours per day for the first 15 days followed by sixteen hours per day for the next 45 days.Seedlings were then transferred to the medium of 1/2 MS+NAA 0.2 mg/L+potato juice 30 g/L+AC 0.5 g/L for plantlets development,after 180 days,the plantlets could be transplanted.
