Abstract

In order to study the influencing factors of rooting culture and to improve rooting ratio,1/4MS was used as basic medium to study the effects of IBA and NAA,sand and agar,sugar and sucrose,distilled water and tap water on rooting culture of Amelanchier alnifolia Nutt.tissue culturing seedlings.Transplantation experiment was also carried out.The results showed that,IBA was not suitable for rooting culture of this plant,and highest rooting ratio was achieved under 1/4MS adding 0.1 mg/L NAA,with 3.59 and 3.57 cm of mean number and mean length of root respectively,and the first class seedlings percentage reached to 64%;sand,sugar and tap water could not be used for rooting culture;the survival ratio of first class seedlings after adapting culture was 98% in the substrate composed of vermiculite and humus with the volume ratio of 1∶2.
