Forsythiaside A inhibit inflammation factor expression on bursa of Fabricius in chicken stimulated by Lipopolysaccharide

Abstract
In order to study the inflammation factor expression that forsythiaside A inhibited LPS on bursa of Fabricius of chicken, forty chicken were randomly divided into control group, LPS group and forsythiaside A(30, 60 mg· kg- 1) therapy group,10 chicken in each group. All chicken were administerated with vehicle, until chicken were 15 d old and were oral administrated by forsythiaside A(30, 60 mg· kg-1) once a day for 7 d on forsythiaside A therapy group. when chicken were 21 d old, after 2 h, when forsythiaside A was oral administrated for last time, and chicken on other three groups except the control group wereinjected intravenously with LPS at 200 μg · kg- 1, and 2 h later, then bursa of Fabricius of chicken were separated for paraffin section and the expression level of mRNA and protein of inflammation factor of bursa of Fabricius in chicken was tested by Real-time PCR and ELISA methods. The results showed that IL-8, IL-17 and IL- 10 mRNA expression and IL- 17 protein content increase and pathological changes appear obviously on bursa of Fabricius of chicken in LPS group, while it is opposite in forsythiaside A(30, 60 mg· kg-1)therapy group, and the above results showed that LPS could increase inflammation factor mRNA expression and protein content, while Forsythiaside A decrease inflammation factor mRNA expression and protein content. It was speculated that Forsythiaside A could inhibit transcription of IL-8, IL-17 and IL-10, interfere with the translation of IL- 17, further affected NF- ?B signaling pathway, and led to inflammation factor release, at last function anti-inflammatory efficacy.
