Effects of He-Ne Laser Irradiation on Bacillus Licheniformis of Producing γ-Polyglutamic Acid

Abstract
Bacillus licheniformis ATCC9945A was irradiated by He-Ne laser. The influences of He-Ne irradiation on the divistion and growth of the bacterium was investigated. The optimum He-Ne laser irradiation dosages was 0.48 mw·cm-2,15 min. Among the two ways of He-Ne laser irradiation,“physiological salt water bacterial suspension liguid” showed more effective inactivation. The bacterial cells in lag phase were the most sensitive to He-Ne laser irradiation,and cells sensitivities decreased gradually in other phases. Thus the bacterial spores were not sensitive to He-Ne laser irradiation. Based on the analysis of the γ-Polyglumatic acid fermentation and electron microscope screening,it was found that He-Ne laser could give rise to an obvious biological stimulating effect and an available mutagenesis effect, several variation strains that produced γ-polyglumatic acid abnormally were screened. In the meantime,by the contrast of PGA、RNA、DNA from the extracell,intracell and periplasmic between mutant strain and original strain,we had a furthe evidence that He-Ne laser can cause mutagenesis effect on Bacillus licheniformis.
