AIM:To establish a revered-phase HPLC method for studying pharmacokinetics of loganin in serum of rat after perfusion.METHODS:The Kromasil C18(4.6 mm × 250 mm,5 μm)column was used as the analytical column and the mobile phase consisted of 0.05% phosphoric acid solution and acetonitrile,with the flow rate at 1.0 mL/min).The UV detection was set at 236 nm.RESULTS:The calibration curves of loganin was linear in the range of 26 ~ 420 ng(r = 0.999 1).The recovery and relative standard deviation of loganin was 94.20% and 4.01%,respectively.The loganin-containing serum was not changed in 24 hours(relative standard 0.55%).After a perfusion of Liuweidihuang Concentrated Pill,the mean plasma concentrations-time relationship was found to fit a one-compartment open model,the main pharmacokinetic parameters of loganin were as follows:Ka,Ke,t1/2Ka,t1/2Ke,Tpeak,Cmax,AUC were 0.008 5 /min,0.025 /min,27.95 min,81.23 min,65.59 min,6 970.40 ng/mL,1 429 620.62 ng/(mL·min).CONCLUSION:This is for the first time to establish the revered-phase HPLC method to determine concentrations of loganin in serum of Liuweidihuang Concentrated Pill and to obtain the pharmacokinetic parameters and characteristics.The method is reliable,the result is accurate.
