Objective To study the effect of sodium fluoride on the collagen in rats in growth period.Methods Ninety 2-month-old SPF Sparague-Dawley rats,half male and female,were randomly divided into 9 groups:control(childhood,adult) group and drug-administered group(childhood,adult high-fluoride and low-fluoride group,long-term high-fluoride and low-fluoride group).The control group were administered orally with solution of 0.9% NaCl,while the drug delivery group were given orally with different dose NaF by the corresponding time.At the experimental endpoint,rats were sacrificed by cardiac puncture under sodium pentobarbital anesthesia.The blood plasma was used to test the content of PICP and ICTP.The serum was applied to test the contents of hydroxyproline.Results ①Compared with CS group,the PICP of the CHS group and CLS group was increased by 38.1% and 38.4%(p0.05) respectively.The change of ICTP of the CHS group and CLS group had not meet statistical significance.The Hyp of the CHS group and CLS group was decreased by 45.7% and 38.6%(p0.05) respectively.②Compared with NS group,the PICP of HS group was losed by30.2%.③Compared with HS group,the PICP of the LS group was added 64.86%,but the Hyp of the LS group was dropped 26.6%(p0.05).The ICTP in LS group had no obvious change.Compared with HS group,the PICP of CHS group were added by 100%(p0.05),and the ICTP in LS group had no obvious change,too.④Compared with NS group,the Hyp of HS group was losed by 47.3%(p0.05),and the Hyp in LS group had no obvious change.Conclusion The long-term use of sodium fluoride can lead to inhibit the synthesis of PICP in rats in growth period,resulting in the loss of bone collagen in rats in growth period.
