Abstract
RÉSUMÉ: Le but de cette recherche est d'explorer la possibilité d'utiliser un pigment naturel pour contrôler la prolifération de bactéries pathogènes en écloserie de bivalves. Le pigment, appelé "marennine", est produit en laboratoire par la microalgue marine Haslea ostrearia. Les modèles biologiques sont les larves de moule bleue Mytilus edulis et la bactérie pathogène Vibrio splendidus. L'hypothèse testée dans le cadre de ce travail est la suivante: la pathogénécité de Vibrio splendidus dans un élevage larvaire de Mytilus edulis peut être contrôlée par l'utilisation de la marennine. L'effet de différentes concentrations de marennine sur la croissance des algues fourrages utilisées pour l'alimentation des larves de moules a été premièrement testé sans démontrer d'influence négative. Ensuite, l'effet des mêmes concentrations de marennine sur la survie, la condition physiologique (analyses lipidiques) des larves et le développement bactérien dans les cultures larvaires a également été testé. Les résultats ont démontré qu'à faible dose, la marennine n'avait pas d' effet cytotoxique sur les larves et qu'elle améliorait leur survie. Finalement, la survie des larves et le développement bactérien dans les cultures ont été testés après 72 heures d'élevage en condition d'exposition à la bactérie pathogène. L'exposition à Vibrio a causé une mortalité significative par rapport au contrôle, mais la survie des larves exposées à la marennine et au Vibrio était égale au contrôle, indiquant que la marennnine a complètement inhibé la pathogénicité de V splendidus. Ces résultats confirment l'intérêt d'utiliser la marennine comme agent protecteur naturel en écloserie. -- Mots clés: Haslea ostrearia, Vibrio splendidus, Écloserie, Bivalves, Antibiotique naturel. --ABSTRACT: \nThe possibility of using a natural microalgal pigment as an antibiotic in bivalve hatcheries was explored. The molecule (named marennine) was produced in the laboratory from Haslea ostrearia cultures. Blue mussel Mytilus edulis was used as a model animal and the pathogenic marine bacteria Vibrio splendidus was used to cause larval mortality. The hypothesis tested was: V splendidus's pathogenicity in a Medulis larval rearing can be controlled using marennine. The effect of different marennine concentrations on growth of microalgae used as larval feed was tested to make sure that feed availability would be the same for aU treatments in further experiments. No difference in growth was found. Then, the effect of the same marennine concentrations was tested on larval survival for 20 days. At a low dose (0.1 mg L- 1 ) , survival was 30 % higher than in the control. A higher dose (1.0 mg L- 1 ) caused heavy Iarval mortality. In a third experiment, ten days old larvae were exposed to V splendidus for 72 hours, with or without marennine. The bacteria caused significant mortality when compared to control, and the marelmine treated larvae exposed to the pathogen had a significantly higher survival, similar to the one in control experiments. We concluded that marennine was an interesting molecule for pathogen control in hatcheries as it is active against bacteria at low concentrations and that is significantly enhanced larval survival. -- Keywords: Haslea ostrearia, Vibrio splendidus, Hatchery, Bivalve, Natural Antibiotic.
