HPLC determination of sildenafil and its demethylated metabolite, UK-103,320, in cat plasma with solid-phase extraction for the pharmacokinetic study

Abstract
Une methode d'analyse simple et fiable par chromatographie liquide a haute performance fut developpee pour la determination simultanee du sildenafil et de son metabolite demethyle (UK-103,320) dans le plasma de chat. Une extraction en phase solide avec une cartouche de SepPak C 1 8 fut utilisee apres deproteinisation, et les taux de recuperation de l'extraction furent de plus de 85% pour le sildenafil et UK-103,320. Le coefficient de variation etait de moins de 15% dans le domaine 50-1000 ng/ml pour les sildenafil et UK-103,320, ce qui indique une reproductibilite acceptable pour la methode. Cette methode fut employee pour une etude pharmacocinetique apres l'administration orale de citrate de sildenafil. Une analyse de moment a revele que la demi-vie d'elimination (7.4 ′ 2.1 hr) etait similaire a celles rapportees dans des etudes avec du [ 1 4 C]-sildenafil, et le pourcentage AUC du UK-103,320 sur le sildenafil etait aussi semblable aux resultats d'un rapport precedent. Ces resultats indiquent que cette methode analytique commode est promptement applicable aux etudes pharmaco-cinetiques de sildenafil et UK-103,320.
