Abstract
Introduction. Increasing experimental data reveal the importance of the gut bacteria in mediating behavioral changes. However, clinical evidences are lacking. In one hand, alcohol-dependent subjects (AD) are known to present with depression, anxiety and neuropsychological disorders. On the other hand, heavy and chronic alcohol consumption is suggested to alter the gut microbiota composition. The aim of this study is to analyze the gut-brain axis in AD subjects. Methods. AD subjects were assessed for biological and psychological parameters. Gut permeability was measured using 51Cr-EDTA. Fecal samples were collected to analyze the gut microbiota composition (using pyrosequencing and qPCR) and function (metabolomic analysis using GC/MS). The inflammatory pathways that were stimulated by gut-derived bacterial products were analyzed in peripheral blood mononuclear cells. Psychological (depression, anxiety, craving) and neuropsychological (cognitive and executive functions) symptoms were assessed by using self-reported questionnaires and neuropsychological tests. The analyses were performed twice in AD subjects, at the first day of alcohol withdrawal and after 18 days of abstinence, and once in healthy controls.
