P3‐040: Design, Synthesis, and Evaluation of Novel Phenoselenazine (PSZ) Based Tricyclic Compounds as Potential Agents to Treat Alzheimer’s Disease

Abstract
Phenoselenazine derivatives were developed to inhibit the aggregation of beta-amyloid, with the ability to simultaneously scavenge free radicals, as potential agents to treat Alzheimer's disease (AD). Synthesis of phenoselenazine derivatives was achieved by coupling substituted diphenylamines with selenium/selenium dioxide. Anti-amyloid aggregation activity was determined by thioflavin T based fluorescence spectroscopy, whereas antioxidant activity was measured based on 2,2-diphenylpicrylhydrazyl scavenging. A group of tricyclic phenoselenazine derivatives were synthesized and evaluated for potential activity toward beta-amyloid aggregation inhibition. Some novel molecules with anti-amyloid aggregation activity and antioxidant properties were identified. Molecular modeling studies were used to understand the binding interactions of these molecules with beta-amyloid aggregates. We designed a new class of tricyclic rings based on a phenoselenazine template as anti-amyloid agents. It is expected that further studies will provide some lead candidates as potential agents to treat AD.
