THE EFFECTS OF DAILY INJECTIONS OF oFSH BETWEEN TWO ENDOGENOUS FSH PEAKS ON FOLLICULAR WAVE DYNAMICS IN ANESTROUS EWES

Abstract
In the ewe, ovarian antral follicles grow in a wave-like pattern. A wave is defined as 1 to 3 antral follicles that emerge or grow from a pool of follicles ≥1 and ≤3 mm in diameter and that grow to ≥5 mm in diameter. Every 4–5 days, a peak in serum follicle stimulating hormone (FSH) concentrations stimulates the emergence of a follicle wave. Previous studies in our lab have shown that exogenous FSH given in the middle of the interwave interval will induce a follicle wave without disrupting endogenous waves. The objective of this experiment was to give daily FSH injections to see if the ovary could respond with daily follicular waves. Transrectal ultrasonography was performed on 12 Western White Face ewes. Six ewes were given 2 injections of ovine FSH (oFSH; 0.35 μg/kg; sc; 8 h apart) daily for 4 days starting 84 h after the detection of a growing 4 mm follicle. The oFSH was prepared in saline with 50% polyvinyl pyrrolidone and 0.05% BSA. The start of treatment coincided with the day after the emergence of an endogenous wave and resulted in serum FSH concentrations similar in magnitude to an endogenous FSH peak. The exogenous oFSH was given between 2 consecutive endogenous FSH peaks. Six ewes received vehicle only. Blood samples were collected daily throughout the experimental period and every 6 h from 24 h before to 5 days after the first injection of FSH. During the 4-day treatment period, mean serum FSH concentrations were higher in the oFSH treated ewes than in control ewes (3.35±0.08 ng/mL, 1.58±0.03 ng/mL; P<0.05). Ewes given oFSH had higher basal serum concentrations of FSH (2.63±0.18 ng/mL, 1.13±0.12 ng/mL) and peak amplitude (0.92±0.07 ng/mL, 0.49±0.04 ng/mL) than control ewes (P<0.05). Daily administration of oFSH induced a mean of 3.8±0.2 waves per ewe with all but 1 ewe showing 4 waves. Control ewes had only 1.0±0.0 wave emerge at the expected time of the next endogenously induced follicular wave. During the treatment period, the number of small follicles (1–3mm in diameter) and serum estradiol concentrations did not differ significantly between oFSH treated and control ewes. However, the number of medium (4mm) and large (≥5mm) size follicles were significantly higher in treatment animals (P< 0.05). Three ewes ovulated 5.3±0.1 days following the first oFSH treatment; however, only 1 ewe formed functional corpora lutea. We concluded that the ovine ovary is capable of responding to daily injections of oFSH with a new follicle wave emerging each day. (poster)
