Einfluss von Promoter-Varianten auf die Genexpression und Plasmaspiegel von Adiponectin und Interleukin-6

Abstract
Adiponectin und Interleukin-6 sind Sekretionsprodukte von Adipocyten, die für den Lipid- und Glucosestoffwechsel, aber auch für chronische adipositas-assoziierte Entzündungsprozesse von Bedeutung sind. Eine Verbindung zwischen Adipositas und Insulinresistenz, Diabetes mellitus und Atherosklerose ergibt sich aus den erhöhten Plasmaspiegeln des proinflammatorischen Interleukin-6 und der erniedrigten Plasmaspiegel des antiinflammatorischen Adiponectins. In einer Studie an 77 postmenopausalen Frauen untersuchten wir die Genexpression von Interleukin-6 und Adiponectin im subkutan-abdominellen Fettgewebe, bestimmten die Plasmaspiegel beider Proteine und sequenzierten beide Gene zur Bestimmung von insgesamt vier Sequenzvarianten. Für die Promotervariante an Position -174 des Interleukin-6-Gens ergab sich folgende Verteilung der Genotypen: 20% CC, 61% GC und 19% GG. Dabei waren weder die Genexpression noch die Plasmaspiegel von Interleukin-6 signifikant unterschiedlich zwischen den Genotypen. Für die Promotervariante an Position -11377 des Adiponectin-Gens ergab sich folgende Verteilung der Genotypen: 46% CC und 54% GC+GG. Hier war die Genexpression in der GC+GG Gruppe größer als in der CC Gruppe (28±20 arbitrary units vs. 13±9 arbitrary units, p<0.001), die Plasmaspiegel unterschieden sich jedoch nicht. Für die Promotervariante an Position -11391 des Adiponectin-Gens ergab sich folgende Verteilung der Genotypen: 85% GG und 15% AA+AG. Weder die Genexpression noch die Plasmaspiegel von Adiponectin waren signifikant unterschiedlich zwischen den Genotypen. Für die Prolin/Alanin-Variante in Codon 12 des Adiponectin-Gens ergab sich ebenfalls keine Unterschied in Genexpression oder Plasmaspiegeln zwischen den Genotypen. Weitere Untersuchungen müssen die Rolle der hier untersuchten und anderer Sequenzvarianten von Interleukin-6 und Adiponectin für die Bildung dieser Moleküle klären.
