The Effects of Polyquad-Preserved Travoprost, Benzalkonium Chloride-Preserved Latanoprost, Purite-Preserved Brimonidine and Preservative-Free Tafluprost on Ocular Surface: A Rabbit Ocular Surface Study

Abstract
Amaç: Farklı prezervan maddeler içeren antiglokomatöz damlalar ile prezervansız damlanın tavşan oküler yüzeyi üzerindeki sitotoksik etkilerinin incelenmesidir. Gereç ve Yöntemler: Yirmi beş adet erkek erişkin Yeni Zelanda beyaz tavşan 5 gruba ayrıldı. Gruplara izotonik %0,9 solüsyon (serum fizyolojik), prezervan içermeyen tafluprost %0,0015 (Saflutan®), %0,15 Purite içeren brimonidin %0,1 (Alphagan P®), %0,001 Polikuad içeren travoprost %0,004 (Travatan®) ve %0,02 BAK içeren latanoprost %0,005 (Xalatan ®) tedavi dozunda 28 gün süreyle uygulandı. Enükleasyon yapıldıktan sonra kornea dokusu ve konjonktiva goblet hücreleri ışık mikroskopisiyle incelendi. Kornea epiteli ve konjonktiva ilişkili lenfoid dokuda (CALT) inflamasyon ve apoptoz bulguları immünohistokimyasal olarak araştırıldı. Sonuçlar ortalama±standart sapma olarak belirtildi. Gruplar arasındaki karşılaştırmalar için One-way ANOVA testi kullanıldı. p<0,05 anlamlı olarak kabul edildi. İstatistiksel analiz GraphPad programı kullanılarak yapıldı. Bulgular: Işık mikroskopisinde, BAK-latanoprost grubunda kornea epitel değişiklikleri ve stroma ödemi daha belirgindi. BAK-latanoprost grubundaki goblet hücre sayısı kontrol, Polikuad-travoprost ve prezervan içermeyen tafluprost grupları ile karşılaştırıldığında anlamlı olarak düşük bulundu (p<0,05). Prezervan içermeyen tafluprost grubunda goblet hücre sayısının diğer gruplara göre yüksek olduğu görüldü (p<0,05). İmmünohistokimyasal incelemede, kornea epiteli ve CALT'da inflamatuar hücre sayısı, prezervan içeren tüm gruplarda anlamlı derecede yüksek bulundu (p<0,05). BAK-latanoprost ve Purite-brimonidin gruplarında apoptotik hücre sayısında artış izlendi (p<0,05). Sonuç: Bu bulgular, prezervan maddelerin kornea epitelinde toksisiteye, konjonktivada goblet hücre sayısında azalmaya, kornea ve CALT'da inflamatuar reaksiyon artışına ve apoptotik değişikliklere neden olabileceğini göstermektedir.
