Einfluß von Stickoxid (NO) auf die Proliferation von Knochenmarkszellen im Sepsismodel der Maus

Abstract
Das in dieser Arbeit zur Sepsisimitation verwendete in-vivo Modell der CASP-Peritonitis bewirkt deutliche Veränderungen in der Knochenmarksfunktion. Die Dynamik der Hämatopoese ist während des septischen Geschehens v.a. durch die Zytokine TNF-a, IFN-g, IL-1b, dem bakterielen Entotoxin LPS und das vermehrt synthetisierte NO empfindlich gestört. Das niedermolekulare NO bewirkt eine Suppression, der durch GM-CSF stimulierten Proliferation im Knochenmark. Die basale Proliferationsrate wird aber dabei nicht verändert. Die Knochenmakrssuppression entsteht durch die direkte und indirekte NO-Wirkung. Die endogene, induzierbare Synthese von NO kann durch spezielle NO- Synthase- Inhibitoren zum größten Teil rückgängig gemacht werden. Die NO-Synthese konnte während der hochakuten initialen Phase der Sepsis als negativer feed- back- Mechanismus gewertet werden um die unkontrollierte Proliferationssteigerung und inadäquate Differenzierung v.a. der weißen Blutzellen zu begrenzen. Die Therapie mit NO- Synthese- Inhibitoren wird aufgrund dieser erbrachten Ergebnisse zusätzlich zur systemisch- Speziellen Antibiose, eine zukunftsweisende Rolle einnehmen.
