Anthocyanin-rich berry extracts and epigallocatechin gallate reduce the enhanced invasiveness of estrogen-negative human breast carcinoma cells elicited by serum

Abstract
LB-149 Breast cancer is the second leading cause of cancer-related deaths in North American women. Despite the evidence for a protective effects of fruits and vegetables on breast cancer remains inconclusive, it has been suggested that fruit and vegetable intake might provide protection against estrogen-receptor negative (ER)-negative but not (ER)-positive breast cancer. However, neither the mechanisms leading to this specificity nor the possibility that a polyphenol-rich intake may protect patient with ER--BC against metastasis have been investigated. In the US, the daily intake of anthocyanin in man has beenestimated to be about 200 mg/day. ER_BC cells (ER--BCC) have markedly higher capacity to migrate, invade and proliferate at site of metastasis than ER-positive one’s. The goal of this study was to determine whether anthocyanin-rich extracts (AREs) inhibit the enhanced motility of highly invasive estrogen-receptor-alpha-negative-MDA-MB-231 BCC elicited by the chemotactic agents found in serum Methods: The cell motility was measured in microchemotaxis chambers containing gelatin pre-coated micropore filters (8-mm pore size). After adhesion to the matrix, the cells were treated with the solvent or with epigallocatechin gallate (EGCg), a laboratory-made red grape skin extract (SGE), a highly bioavailable bilberry extract or pilot red grape skin, blackberry, fermented-grape or black currant extracts. The cell migration was initiated by FBS (0.5%) and measured 5 hrs after cell elicitation. Results As expected, serum-stimulation elicited strong MDA-MB-231 cell motility. Although at 25 µg/mL, all tested AREs reduced this chemotactic effect, their respective potency was markedly different. Grape and bilberry AREs provided the highest protection. Both exhibited dose-dependent inhibitory effect. At 25 µg/mL, the most potent one (SGE) decreased the motility of FBS-stimulated MDA-MB-231 cells by up 80% and, was found to be, respectively 3.2- and 2-fold more potent than EGCg and the bilberry extract at similar concentrations. Conclusion. This is the first demonstration that in vitro, anthocyanin-rich berry extracts are able to inhibit the migration and invasiveness of ER--BCC. This suggests that a regular consumption of berries (especially grape and bilberry) might provide protection against the development of primary and secondary ER--breast tumors. New experiences will test this hypothesis in animal model.
