Abstract
Background: Epidemiological studies demonstrate an excess of circulation diseases in different groups of exposed to radiation (A-bomb survivors, Chernobyl cleanup workers etc.). Whether post-radiation heart disease have a distinct molecular profile is not clear. Aim of the study was to estimate the role of gene regulation of cell cycle and telomere length in coronary artery disease. Patients. Subjects (n-160) were selected randomly among the Chernobyl cleanup workers cohort. Methods: Relative telomere length (RTL) was studied by Flow-FISH assay with Telomere PNA Kit. Cellular phenotype, p53, bcl2 regulatory proteins expression and apoptosis in Annexin-V test, CD95+ fraction were studied by flow cytometry, TERT, TRF1, TRF2, TP53 gene expression in peripheral blood leukocytes by RT-PCR. Results: In patients with chronic coronary disease of I and II functional class an increased TP53 and TP53I3 genes expression was associated with increased CD95+ fraction, lower BAX expression and positive correlation between CCND1 gene and bcl-2 expression and radiation dose. Highest expression of CCND1 gene was observed in vascular dementia. The telomere length was reduced after 50 years (6.1%). The most significant reduction in telomere length was shown after 70 (11.7%) and in exposed in earlier age (before 25). In FC III coronary artery disease the TRF1 and TRF2 genes over-expression was associated with TERT down-regulation and low RTL.
