Pada lima tahun terakhir ini, para klinisi dibeberapa negara mengamati
\nadanya kasus deman tifoid anak yang berat bahkan fatal, yang ternyata disebabkan
\noleh strain S. typhi yang telah resisten terhadap antibiotik yang lazim
\ndipergunakan untuk pengobatan demam tifoid. Strain yang telah resisten terhadap
\ndua atau lebih jenis antibiotik yang lazim dipergunakan yaitu ampisilin,
\nkloramfenikol, dan kotrimoksazol dinamai strain Multi Drug Resistance (MDR)
\n(Hadinegoro, 1999).
\nAda beberapa gen target dan metode berbeda yang digunakan untuk
\nmendeteksi Salmonella spp antara lain multiplex PCR pada rfbE (gen terkait
\naktivitas rantai samping lipopolisakarida), fliC (gen pada sintesis flagella), invA
\n(gen pada invasi sel/jaringan), spvC (gen virulen plasmid Salmonella), sebagai
\ngen target, uniplex PCR pada gen rRNA dan nested-PCR untuk gen flagel
\n(Banavandi, et al., 2005; University of Calgary, 2002). Salmonella enterotoxin
\ngene (stn) disekuensi dari serotip Salmonella enterik: Typhimurium, Typhi,
\nParatyphi A dan B. Sekuens dari keempat serotip menunjukkan homolog dengan
\nsekuens gen stn dari serotip Typhimurium yang telah dilaporkan (Hassan, Verma,
\nNabi, 2004).
\nTeknik PCR telah menyuguhkan peningkatan kepekaan, menghasilkan
\nwaktu pemrosesan yang lebih cepat dan meningkatkan kemungkinan
\nterdeteksinya bakteri patogen. Sebagai tambahan dalam analisis makanan, PCR
\ntelah sukses digunakan dalam mendeteksi dan mengidentifikasi organisme
\npatogen dari sampel klinis dan lingkungan (Hassan, et al., 2004).
\nPada penelitian ini dilakukan uji aktivitas biokimia bakteri Salmonella spp
\ndengan Triple Sugar Iron Agar (TSIA), menggunakan metode Polymerase Chain
\nReaction (PCR) untuk mendeteksi gen spesifiknya, dimana gen spesifik yang akan
\ndideteksi adalah gen stn, dan melakukan uji sensitivitas bakteri terhadap beberapa
\njenis antibiotika. Setelah penelitian ini dilaksanakan diharapkan dapat mengetahui
\nsifat biokimia Salmonella spp pada media TSIA, mendeteksi keberadaan gen
\nspesifiknya, dan mengetahui sensitivitasnya terhadap antiobiotik uji.
