Abstract

Microcystinは淡水藍藻の一部の種が産生する非リボソ―ムペプチドであり、強力な肝臓毒として知られている。このMicrocystin生合成のためにはD-glutamate と D-MeAsp/D-aspartateをペプチド骨格に組み込むラセマーゼ活性が必要となる。本研究ではmicrocystin合成酵素遺伝子mcyクラスターに位置するラセマーゼ遺伝子mcyFに着目し、microcystin産生藍藻の系統発生について考察を行った。13株のmicrocystin産生藍藻株においてmcyFホモログ遺伝子が確認され、McyFはmicrocystin生合成に関連する酵素と言える。9株のMcyFのアミノ酸解析ならびに系統発生解析を行った。保存されたアミノ酸モチーフの解析結果からMcyFはglutamate racemase よりaspartate racemaseの機能をすることが予想された。系統発生解析の結果から、McyFがaspartate/glutamate racemaseの系統樹において明らかにaspartateのグループに近い位置にあることが判明した。
