The cover feature picture shows how a change in the topology of an optimized genetically encoded Ca2+ indicator, GCaMP6s, led to a variant that retains the favorable properties of the original but also has new termini in closer proximity to the chromophore. Fusion of a bright luciferase to GCaMP6s (with the original topology) does not generate high bioluminescence resonance energy transfer (BRET) efficiency between the donor and acceptor either in the presence or absence of Ca2+. However, fusion of bright luciferase to the topological variant of GCaMP6s results in high BRET efficiency in the presence of Ca2+ and low BRET efficiency in its absence. This fusion protein is thus a highly effective glow-in-the-dark Ca2+ indicator. More information can be found in the communication by R. E.
