The cover feature picture shows Goldilocks offering a pentanucleotide (flower) to the middle-sized Mama bear who holds the ideal linker length to display the R group modification. In our work, we explored the effects of decreased linker lengths on the efficiency, fidelity, and bias of Ligase-catalyzed OligOnucleotide PolymERizations (LOOPER). Whereas longer linker lengths can have a negative impact on binding thermodynamics, shorter linker lengths result in high error and bias during LOOPER. An ethane-1,2-diamine linker was found to be optimal in all respects, and should facilitate the discovery of highly modified aptamer isolated from in vitro evolution of LOOPER-derived DNA libraries.
