Abstract
グリコデンドリマー研究の最近10年は、グリコデンドリマーのデザインと生物学的応用の両方の点で、非常に大きく洗練されてきたことが証明されている。これらの人工的な多分岐糖鎖の中には動物試験にまでたどり着いたものもあり、その結果は、特に、細菌の接着や異種移殖に関係するガリリ (Galili) 抗原を用いて観察された抗体の中和などで非常に有望である。鍵となる基本構造が大量でも市販で入手できるようになり、いくつかのデンドリマーが細胞や動物でのアッセイ系で無毒であることが示されている。この美しい構造は多価の糖鎖-タンパク質相互作用に新たな光を注ぐことを可能にした。事実、多価もしくは凝集したタンパク質受容体を交差結合させる能力は、古典的なグリコシドクラスター効果を再定義するのに役立っている。多価効果を、Nostoc ellipsosprum から単離された高親和性オリゴマンノース結合タンパク質である cyanovirin を用いた最近の研究例によって改めて議論する。グリコデンドリマーの有利な点をグリコポリマーとの比較で簡単に議論し、続いて合成戦略やこれらの物質に調製に使われた共通の基本構造について詳細に述べる。グリコデンドリマーの構造的な欠点を述べ、立体的な隠蔽とそれに寄因する高濃度条件での糖の接近しにくさに伴う問題点を解決に導く方法を提示する。また最も一般的な化学的結合法や化学酵素的な合成についても概説する。最後に、パラジウムやコバルトを用いる遷移金属触媒を用いた交差結合反応による糖の共有結合の迅速な構築を含む、新世代の「スマート」なグリコデンドリマーやグリコクラスターの合成も述べる。動的なコンビナトリアルケミストリー (アダプティブケミストリー) によるグリコデンドリマーライブラリーの芸術的な調製や、遷移金属の回りの自己組織化が、より創造的なレベルの分野を切り開いている。いくつかのタンパク質受容体 (例えばガレクチン) が、価数や3次元の位相に基づいて選択した同じ糖鎖リガンドの結合を予測できる事実によって、新しい戦略が特に興味が持たれている。
