WND1B gene from Populus trichocarpa is homologous to SND1,which is a key regulator of secondary wall synthesis in fiber cells of Arabidopsis.The 5' regulatory region of WND1B initiation codon advanced position about 2100 bp was cloned by PCR.TATA box,CAAT box and other core configurations were found in this promoter region.Several sequences similar to eukaryotic cis-regulatory elements were found in the 5'UTR proximal 5' flanking sequence.A series of truncated sequences of WND1B promoter were generated.The expression vectors pCAMBIA1301POG-1BP/1BP-F2/1BP-F3/1BP-F4-GUS were constructed and the transient expression analysis was carried out.The GUS activity determination revealed that the fragment of PtrWND1BP/ 1BP-F3/1BP-F4 could drive expression of the GUS gene while PtrWND1BP-F2 could not.The results suggested that the 5'UTR of PtrWND1B affected for the promoter activity.
