P3‐257: ASSOCIATION OF PLASMA NEUROFILAMENT LIGHT AND WHITE MATTER DENSITY IN ALZHEIMER'S DISEASE

Abstract
There is increasing interest in plasma neurofilament light (NfL) as a potential biomarker for axonal injury in different neurodegenerative diseases, including Alzheimer's disease (AD). A few studies have shown that cerebrospinal fluid and plasma NfL levels correlate with white matter (WM) loss in several brain regions. Here we aim to investigate this association in the McGill Translational Biomarkers in Aging and Dementia (TRIAD) cohort, a specialized deeply phenotyped cohort. WM Voxel-based morphometry (VBM) of 133 subjects (105 cognitive unimpaired (CU); 28 AD) was generated using Dartel (8mm FWHM) and spatially normalized to the ADNI template. NfL plasma levels were measured using an in-house Single molecule array (Simoa) method. Linear models were performed to test the association between NfL and VBM as well as to compare the two groups, correcting for age, gender and the time difference between the measurements. Results show a negative correlation between NfL and WM density. The differences were observed in the corpus callosum and frontal and parietal white matter in CU. In AD, this association is confined to the precuneus WM as well as in the frontal and inferior temporal lobe WM bilaterally. The statistical difference between groups was seen in the inferior temporal lobes and in frontal WM.
