O2‐11‐05: MILD BEHAVIORAL IMPAIRMENT IS ASSOCIATED WITH β‐AMYLOID BUT NOT WITH TAU IN COGNITIVELY INTACT ELDERLY INDIVIDUALS

Abstract
Mild behavioral impairment (MBI) is a neurobehavioral syndrome characterized by the emergence of sustained non-cognitive neuropsychiatric symptoms in elderly persons. MBI therefore represents a potential marker of preclinical Alzheimer's disease. While studies have revealed correlations between certain neuropsychiatric symptoms and Alzheimer's disease (AD) biomarkers, little is known about the association of MBI with brain β-amyloid pathology. Aim: to investigate whether mild behavioral impairment is associated with accumulation of β-amyloid in cognitively normal elderly persons. Seventy-one cognitively healthy individuals aged from 57 to 86 years old underwent neuropsychological evaluation as well as MRI, [F]-MK6240 tau PET and [F]-AZD4694 β-amyloid PET. [F]-MK6240 and [F]-AZD4694 standardized uptake value ratios (SUVRs) used cerebellum grey matter as the reference region and were calculated between 90 to 110 min and 40 to 70 min post-injection, respectively. MBI was assessed using the Mild Behavioral Impairment Checklist (MBI-C) which is composed of 5 domains: decreased motivation, emotional dysregulation, impulse dyscontrol, social inappropriateness, and abnormal perception. Voxel-based regressions were used to evaluate the relationship between [F]-AZD4694 and [F]-MK6240 retention and MBI-C scores, adjusting for age, gender, and education, and APOE status. All voxel-based regression analyses were corrected for multiple comparisons using random field theory at p < 0.001. A positive correlation was found between total MBI-C scores and [F]-AZD4694 retention in the frontal and temporal lobe, as well as in the basal ganglia. The MBI-C domain scores for decreased motivation, emotional dysregulation, and impulse dyscontrol were highly correlated with [F]-AZD4694 retention in the left frontal lobe and basal ganglia, while abnormal perception scores were found to be correlated with [F]-AZD4694 retention in the right frontal lobe as well as in the occipital lobe. No correlation was found between [F]-MK6240 retention and MBI-C scores.
