Abstract
Adiponectin is an adipocyte‐derived peptide hormone involved in energy homeostasis and the pathogenesis of obesity, including hypertension. We have identified potential roles for adiponectin in controlling the excitability of single neurons in important autonomic control centres known to play important roles in the control of feeding and metabolism. The area postrema (AP) which lacks a blood brain barrier is a critical homeostatic integration centre for humoral and neural signals which expresses mRNA for both AdipoR1 and AdipoR2 receptor. Microinjection of adiponectin into AP caused significant increases in arterial blood pressure. Patch clamp recordings from AP neurons show that 60% of these cells are sensitive to adiponectin. Furthermore single cell recordings coupled with single‐cell RT‐PCR indicate that adiponectin responsive AP neurons express both subtypes of receptor. Within the hypothalamus the paraventricular nucleus has also been suggested to represent an important potential target at which adiponectin may act to influence feeding and metabolism. Single cell recordings from PVN neurons show specific effects of adiponectin of oxytocin (hyperpolarization) and vasopressin (depolarization or hyperpolarization) secreting neurohypophysial neurons. These data suggest important roles for adiponectin in controlling the excitability on CNS neurons involved in the control of feeding and metabolism.
