Nutritional Science has much paid attention to the well-being of the human gut recently because of the increase in health problems associated with the human gut. There is a much demand in prebiotics today, which are essential for maintaining a healthy nature in human gut. Prebiotic is a non-viable food ingredient, selectively metabolized by the healthy gut bacteria such as Lactobacillus and Bifidobacterium. The focus of this research was to determine the potential prebiotic activity using the methanol polyphenolic extractions of Fragaria ananassa (Strawberry) and Vitis vinifera (Grapes), to investigate the growth of Lactobacillus acidophilus LA-5 which was directly obtained. Probiotic potential of the isolate was determined using viable cell count technique and further the availability of the phytochemicals of the raw materials were assessed using qualitative phytochemical analysis. The extracts were introduced to De Man, Rogosa and Sharpe agar (MRS agar) medium separately and were inoculated with Lactobacillus acidophilus LA-5 and incubated at 370C for 48 hours. In the positive control, culture (1 x 10-6 CFU/ml) was grown in fresh MRS broth enriched with 0.05% cysteine hydrochloric omitting prebiotic ingredients. Phytochemical analysis on saponins, anthocyanin, coumarin, flavonoids, terpenoids and xanthoprotein were determined qualitatively. The results revealed that polyphenolic extracts obtained from fresh F. ananassa have a desirable prebiotic activity (6.25 CFU/ml + 0.00577) compared to that of other extracts (100% Strawberry/ Dried Sample - 6.20 CFU/ml + 0.0252, 100% Grapes/ Dried Sample - 6.23CFU/ml + 0.01528, 100% Grapes/ Fresh Sample - 6.21CFU/ml + 0.01528). Moreover, F. ananassa is able to promote the growth of L. acidophilus than the positive control in a statistically significant (p< 0.05) manner. In qualitative screening, saponins, anthocyanin, coumarin, flavonoids, terpenoids and xanthoprotein were detected in higher intensities in F. ananassa. These findings illustrate that F. ananassa have desirable prebiotic properties on L. acidophilus LA-5. This provides an opportunity for further investigation of Fragaria ananassa for its prebiotic activity and quantitative compound screening.
