Regulation of early stages in clathrin mediated endocytosis revealed by quantitative analyses in living cells

Abstract
Clathrin‐mediated endocytosis (CME), which occurs at clathrin coated pits (CCPs), requires the major coat proteins, clathrin and AP2 complexes together with a myriad of endocytic accessory factors and lipids. CME proceeds through multiple stages, including initiation, stabilization of nascent CCPs, and maturation during which CCPs collect cargo molecules, grow in size and increasingly invaginate until they remain connected to the cell surface via a narrow neck. The GTPase dynamin assembles onto this neck and drives membrane fission to release clathrin coated vesicles and complete the process. Total internal reflection fluorescence microscopy (TIR‐FM) in living cells expression fluorescently‐tagged coat proteins is a powerful tool to study the assembly, maturation and dynamic behavior of CCPs; however, extracting reliable measurements from these TIR‐FM movies represents a major challenge. We have developed new methods for particle detection and tracking that enable accurate, unbiased and global analyses of CCP dynamics even at low signal:noise. These assays have revealed striking phenotypes resulting from the expression of mutant AP2 adaptors that could not be detected using conventional biochemical assays for CME.
