MLCP activation limits oscillatory pulse pressure‐induced rise in myogenic tone of isolated mouse cerebral arteries in the absence of NO

Abstract
In response to a static luminal pressure (SP), cerebral resistance arteries develop myogenic tone (MT) in vitro that is magnified by eNOS inhibition. The natural stimulus of MT, however, is the oscillatory pulse pressure (PP) induced by the cardiac cycle. We studied the impact of a PP (550 bpm) of 30 mm Hg on the MT of pressurized (60 mm Hg) mouse cerebral arteries (~120 μm diameter, n=7 per group) before and after eNOS inhibition. MT was magnified by PP compared to SP (14±2 vs. 6±3% reduction in diameter, p<0.05). As expected, inhibition of eNOS with L‐NNA (10 μM) increased MT in SP from 6±3 to 29±4% (p<0.05); PP, however, did not further increase MT (14±2 and 18±3%, respectively). MT is limited by the myosin light chain phosphatase (MLCP); phosphorylation of its MYPT1 subunit on Thr850 inhibits its activity and was unchanged between SP and PP. In contrast, eNOS inhibition led to a decrease in P‐MYPT1 by PP (−41%, p<0.05), while it was unchanged during SP (+12%, p>0.05). In conclusion, MT is maintained constant after eNOS inhibition under physiological conditions of oscillatory PP through a rise in MLCP activity. Supported by CIHR MOP 89733.
