Coactivator-associated arginine methyltransferase 1 (CARM1) catalyzes the methylation of arginine residues on target proteins, which drives numerous gene expression programs. For example, the expression of two known estrogen-related genes, complement C3 and stanniocalcin 2, are reduced in murine CARM1 knockout embryos, demonstrating a genetic link between estrogen action and CARM1. CARM1 also has important functions during myogenesis, as well as in response to exercise and during disuse-induced phenotypic plasticity. However, a mechanistic understanding of the role(s) of CARM1 in skeletal muscle biology is lacking. PURPOSE: To determine the impact of CARM1 on skeletal muscle biology. Specifically, in this study, we test the hypotheses that 1) Skeletal muscle CARM1 is required to maintain habitual levels of volitional physical activity, and 2) The impact of CARM1 removal on muscle biology will differ between males and females. METHODS: To investigate the role of CARM1 in muscle biology we generated skeletal muscle-specific CARM1 knockout (mKO) mice. Furthermore, we used chronic exercise as a physiological stressor in an attempt to gain additional insight into the function of CARM1 in muscle. At 12 weeks of age male (M) and female (F) wild-type (WT) and mKO littermates were randomly assigned to either sedentary (SED) control or voluntary wheel running (VWR) groups for 8 weeks. Wheel revolutions were measured every 10 minutes and converted to distance traveled (km). Running wheels were removed 24-48 hours before a variety of tissues were collected. Western blotting was performed to confirm animal genotypes in the tibialis anterior (TA) muscle. RESULTS: In total, WT and mKO animals ran a similar distance every day (WT 3.1 ± 0.5 km vs. mKO 2.8 ± 0.3 km) for 8 weeks. However, when examining the running wheel data while accounting for sex, we observed that F WT animals completed a greater volume of exercise per day then mKO animals (WT 5.5 ± 1 km vs. mKO 3.2 ± 0.4 km, p < 0.05), while no difference was observed between M mice (WT 2.2 ± 0.5 km vs. mKO 2.3 ± 0.5 km). VWR, genotype, and sex significantly impacted body mass and muscle mass. CARM1 protein expression in TA muscles was similar between SED and VWR WT animals. CONCLUSIONS: Our data suggest that removal of CARM1 negatively impacts the ability of F mice to exercise.
