Abstract

Objectives This study aimed to evaluate the efficacy of a supplement product containing salmon roe oil-derived DHA and EPA on cognitive function and mental health-related QOL in middle-aged and older adults. Methods A randomized, double-blind, placebo-controlled, parallel-group study was conducted in 100 participants aged 60 to 85 years with subjective memory complaints or symptoms of forgetfulness pointed out by close relatives. Participants were randomly assigned to receive a test product containing salmon roe oil(DHA and EPA)or placebo for 12 weeks. Primary endpoint was evaluated by Japanese version of Montreal Cognitive Assessment (MoCA-J), and secondary endpoints were evaluated by Mini-Mental State Examination (MMSE), 36-Item Short Form Survey(SF-36). Results Significant improvement was observed in both the mental health and the mental component scores(2MCS and 3MCS)of SF-36 in the test product group compared with the placebo group at Week 12. In a post hoc analysis of the MoCA-J scores by exercise habits, significant improvement was observed in a language test score in the subgroup with exercise habits at least once a week and an attention test score in the subgroup without exercise habits in the test product group. No clinically significant adverse events were reported during the study. Conclusions The results suggest that the ingestion of the test product improves(1)mental health-related QOL,(2)verbal memory for people with exercise habits at least once a week, and (3)working memory and attention function for people without exercise habits in middle-aged and older adults.
