Dietary Resveratrol Enhances Dilatory Responses but Does Not Alter eNOS Levels in Aorta of Spontaneously Hypertensive Rats

Abstract

Resveratrol is an active compound found in the skins of grapes which may be responsible for the health benefits of moderate red wine consumption. Spontaneously Hypertensive Rats (SHR) were administered resveratrol in drinking water at 0, 0.448, or 4.48 mg/l for 28 days. The low dosage was chosen to mimic moderate red wine consumption. Following treatment, thoracic aorta rings were excised for in‐vitro assessment of vasomotor function. Chronic resveratrol intake significantly improved endothelium‐dependent relaxation to acetylcholine, increasing maximal values to 80.8±5.2% and 80.8±5.0% in low and high dosage groups compared to 60.7±1.4% in controls (P < 0.01). The effects of resveratrol were abolished in the presence of eNOS inhibitor L‐NAME, as there was an elimination of ACh‐induced dilatation in all groups. Resveratrol did not alter the contractile responses to KCl or phenylephrine, or the relaxing effects of sodium nitroprusside. Immunoblots of thoracic aorta revealed no treatments effects on eNOS, superoxide dimutase 1 and 2, or gp91phox. This data indicates that chronic resveratrol treatment at doses which mimic moderate red wine consumption improves endothelium‐dependent dilation in the SHR.
