Physical Data: mp 272.9 °C, d = 1.115 g/cm−3. Solubility: water, toluene, dichloromethane, acetonitrile, tetrahydrofuran. Form Supplied In: commercially available. Colorless viscous liquid. Analysis of Reagent Purity: 1H NMR, 13C NMR, and 31P NMR.1 Preparative Methods: Two methods have been reported in the literature. The first method involved deprotonation of di-tert butyl phosphate with sodium bicarbonate and tetra-n-butylammonium hydrogen sulfate in water, followed by quenching with a chlorosulfate1 (eq 1). (1) The second method employs phosphorus trichloride as the starting material, which is reacted with tert-butanol in dichloromethane to afford the di-tert-butylphosphite. Subsequent oxidation using potassium iodide/hydrogen peroxide solution afforded di-tert-butylphosphate. Finally, di-tert-butylphosphate is alkylated with chloromethyl chlorosulfate in the presence of dipotassium hydrogen phosphate and a chlorosulfate to afford the desired compound (eq 2)2 (2) Purification: Purified by flash silica gel column chromatography using ethyl acetate/hexanes (1:3). Handling, Storage, and Precautions: tested positive for mutagenicity in the AMES assay. Should be handled accordingly. Use in a fume hood.
