Inflammatory-related Cytokine Responses to Endurance Exercise in Healthy Men following Estrogen Supplementation

Abstract
0906 Prolonged exercise is often associated with an increase in plasma concentration of several inflammatory-related cytokines. We have shown that women exhibit an attenuated inflammatory response to unaccustomed exercise, compared with men (Stupka et al. 2000 J. Appl. Physiol. 89: 2325–2332). Therefore, we hypothesized that the cytokine response to prolonged cycling would be modulated by the female sex hormone estrogen. PURPOSE: To determine the influence of estrogen supplementation (ES) on inflammatory-related cytokine responses to prolonged cycling in healthy men. METHODS: In a randomized, double-blind, cross-over design, nine men (21 ± 1 y, mean ± SD) cycled for 90 min at 65% VO2 max after eight days of either ES (2 mg 17β-estradiol/day) or placebo (PL, polycose). Following a two-week washout period, they repeated the exercise after eight days on the alternate treatment. On each test day, venous blood was drawn prior to and immediately following exercise. Plasma concentrations of interleukin (IL)-1β, IL-6, IL-8, IL-10, IL-12 and tumor necrosis factor-α (TNF-α) were determined by flow cytometry with a cytometric bead array assay (BD Biosciences, Human Inflammation CBA kit). RESULTS: IL-1β and TNF-α were not reliably detectable in the majority of samples. IL-6 increased (p < 0.001) similarly after ES and PL, from 2.5 ± 1.5 pg/mL at rest to 5.8 ± 2.6 pg/mL following exercise. IL-8 increased significantly (p < 0.01) from 5.5 ± 2.5 pg/mL at rest to 8.1 ± 2.8 pg/mL following exercise with ES, and from 5.5 ± 2.0 pg/mL at rest to 9.5 ± 3.8 pg/mL following exercise with PL (interaction NS, p = 0.14) Fineither IL-10 (p = 0.10) nor IL-12 (p = 0.83) increased significantly with exercise, and there was no effect of ES. CONCLUSION: Although 90 min of moderate intensity cycling increased circulating levels of inflammatory-related cytokines in healthy young men, eight days of ES had no significant effect on these changes.
