Abstract
Several dominant avirulence genes from the smut fungus, Ustilago hordei, have been identified on differential cultivars of barley.We analyzed 50 progeny segregating for three unlinked Avr genes, VI, V2, and V6.Two RAPD markers, loosely linked to VI at 7 and 2 map units, have been found by bulk segregant analysis.One AFLP marker, linked to the recessive allele vl, was found physically closer but still at 2 cM distance.A cosmid carrying this AFLP marker harbored a nearby probe, common to both parents, that could not detect recombinants.The area is embedded in a repetitive, difficult-to-clone DNA region.A BAC-library representing the avirulent parent yielded several overlapping clones.Genomic walking identified probes that, upon recombinational analysis, delimited VI on an approximately 40-kb region.Pathogenicity tests with subclones are underway.Only germinating seedlings can be inoculated with mated strains and success rates during compatible interactions vary from several to 80%.Reliable scoring is possible only at heading after 8 weeks.The incompatible interaction does not show any visible, macroscopic symptoms such as lesions or HR.We have also found through phylogenetic analysis of ITS sequences and AFLP fingerprint comparisons, that the small grain-infecting smuts which include U. hordei, consist of a closely related group of organisms pathogenic on related hosts. Molecular characterization of DNA encoding 16S-23S rRNA intergenic spacer regions and 16S rRNA of pectolytic Erwinia species.
