Effects of Linker Rigidity and Orientation of Mannoside Cluster for Multivalent Interactions with Proteins

Abstract
A series of di-, tri-, and hexameric a-D-mannopyranoside clusters having rigid linkers were constructed in order to evaluate the effects of inter-saccharide distances upon multivalent binding interactions with the phytoheamagglutinin from Canavalia ensiformis (Concanavalin A). To this end, O-aryl and O-propargyl glycosides were synthesized and then coupled together using transition metal catalyzed cross-coupling reactions. Thus, ortho-, meta-, and para-iodophenyl mannosides were treated with acetylene or propargyl mannoside under palladium(0)-catalyzed Sonogashira conditions to afford mixtures of cross-coupled symmetrical and unsymmetrical dimers. Dicobalt octacarbonyl benzannulation of acetylenic mannosides provided hexamers showing potent cross-linking ability.
