Abstract
> La voie de signalisation ERK (extracellular signal-regulated kinase) consiste en une squence de protines kinases, RAF/ MEK(MAP/ERK kinase)/ERK, impliques dans l'activation des kinases ERK1 et ERK2 [1, 2]. Chez les mammifres, chaque niveau de cette voie implique plus d'une enzyme. Ainsi, trois protines RAS codes par les gnes paralogues 1 Kras, Hras et Nras peuvent lier et activer les trois protines kinases RAF codes par les gnes Araf, Braf et Raf1. Les protines RAF, une fois actives, peuvent phosphoryler et activer les kinases MEK1 et MEK2, codes par les gnes Mek1 et Mek2. MEK1 et MEK2 sont responsables de la phosphorylation des protines ERK1 et ERK2, codes par les gnes Erk1 et Erk2. MEK1 et MEK2 sont les seules kinases connues capables d'activer ERK1 et ERK2, et elles sont considres, ce titre, comme des cibles thrapeutiques prometteuses, puisque la suractivation des kinases ERK est frquemment observe dans diverses maladies, incluant les cancers et les maladies auto-immunes. La prsence de plusieurs paralogues aux diffrents niveaux de cette voie de signalisation reflte d'ailleurs la complexit du contrle de son activation dans les diffrents processus cellulaires o elle est implique chez les mammifres.
