Caracterização do índice de cobertura por pericitos em neoplasias mamárias caninas e seu papel como fator prognóstico

Abstract
A angiogênese é um fator importante em tumores mamários caninos e está relacionado à malignidade desta neoplasia. Estudos sobre o papel dos pericitos, células importantes para a estrutura vascular, ainda não foram realizados nas neoplasias mamárias desta espécie e podem ser úteis para o desenvolvimento de terapias contra esta enfermidade. Objetivos: Comparar o índice de cobertura por pericitos (PCI) da microvasculatura de diferentes subtipos e graus histológicos de neoplasias mamárias em cadelas e avaliar a relação entre o PCI e a sobrevida dos animais para testar seu valor como fator prognóstico. Metodologia: Foram utilizadas 78 amostras histopatológicas de diferentes subtipos de neoplasias mamárias caninas referentes a 54 animais que foram submetidas à imuno-histoquímica por dupla marcação sequencial utilizando anticorpos primários anti-vWF, para marcação de células endoteliais e determinação da densidade microvascular (MVD), e anti-α-SMA, para identificação de pericitos e determinação do PCI (vWF+, α-SMA+ / vWF+, α-SMA-). Resultados: O PCI de neoplasias mamárias malignas foi significantemente menor (p = 0,0135) que o das benignas (56,93% versus 71,21%), mas não houve diferenças entre as MVDs. Posteriormente, os pacientes foram separados em dois grupos utilizando-se o ponto de corte de PCI = 70,16%. Não foram detectadas diferenças significantes entre as curvas de sobrevida dos grupos PCI>70,16% e PCI<70,16%. Conclusões: As neoplasias mamárias em cadelas apresentam cobertura de pericitos variável, maior nas neoplasias benignas, o que pode indicar maior estabilidade vascular neste grupo. Não foram detectadas diferenças significantes para o PCI entre os graus histológicos das neoplasias malignas e o PCI não foi considerado um bom fator prognóstico para as neoplasias mamárias.
