Changes in plasma anti-inflammatory cytokines, such as interleukin-6 (IL-6) and IL-10, following acute resistance exercise (RE) have been widely explored. However, the relationship between changes in plasma cytokines and inflammatory signaling pathways in immune cells following acute RE is unknown. PURPOSE: This study aimed to explore the relationship between inflammatory signaling pathways in peripheral blood mononuclear cells (PBMCs) and circulating cytokines following acute RE. METHODS: Healthy young males (n = 16; age 23.5 ± 2.7 years, body mass index 22.4 ± 1.7 kg/m2) participated in a randomized crossover trial involving RE and sedentary control (CON). Participants performed RE in the following order: deadlift, bench press, squat, and barbell row (4 sets with 70% 1RM and the last set to failure). Blood samples were collected at the corresponding time points. RESULTS: Elevated circulating IL-6 levels were observed in RE at post 0 (1.25 ± 0.65 vs. 0.76 ± 0.41 pg/mL) and post 4 h (4.31 ± 3.20 vs. 2.21 ± 0.67 pg/mL) compared with CON (p < .05). Higher circulating IL-10 levels were observed in RE than CON at post 0 (1.02 ± 0.29 vs. 0.86 ± 0.26 pg/mL; p < .05). No differences in TNF-ɑ levels were observed between both trials (p < .05). Changes in total STAT3-ɑ and STAT3-β expression in cytoplasmic fraction of PBMCs were not different between both trials (p > .05). Increased phosphorylation of STAT3-ɑ (4.8-fold vs. 1.5-fold) and STAT3-β (3.4-fold vs. 1.3-fold) in cytoplasmic fraction of PBMCs were observed in RE compared with CON at post 4 h (p < .05). No differences in STAT3 and SHIP1 expression in nuclear fraction of PBMCs were observed following RE (p > .05). SOCS3 expression in cytoplasmic fraction of PBMCs was greater in RE than CON at post 0 (1.9-fold vs. 0.8-fold), post 2 h (1.9-fold vs. 0.9-fold) and post 4 h (1.9-fold vs. 1-fold; p < .05). CONCLUSIONS: These results suggest that acute RE-induced IL-6 response coincides with activation of anti-inflammatory STAT3 signaling in PBMCs.
