An In Vitro Pilot Study Investigating the Effect of Activated Autogenous Platelet-Rich Plasma (ALP-PRP) on Osteogenic Differentiation of Equine Bone Marrow-Derived Mesenchymal Stem Cells

Abstract
Introduction : Platelet rich plasma is a biological therapy which is used clinically to promote the healing of musculoskeletal tissues after injury. Little evidence exists for the use of PRP in equine bone healing. We hypothesized that the addition of activated leucocyte poor platelet-rich plasma (ALP-PRP) to equine bone marrow derived mesenchymal stem cell (BMMSC) and osteogenic cell cultures would promote bone production in vitro. Materials and Methods : BMMSC and osteogenic cell cultures were used to evaluate ALP-PRP in promotion of bone healing. Animal study protocols were approved by the Animal Care Committee. The experimental design included four groups (1) BMMSCs in standard media (SM), (2) BMMSCs in SM with PRP, (3) BMMSCs in osteogenic media (OM), and (4) BMMSCs in OM with PRP which were evaluated for proliferation, osteogenic gene expression, and bone nodule production. ANOVA and Tukey's test were used for comparisons among groups. Results : Addition of ALP-PRP to mesenchymal stem cell cultures and osteogenic cell cultures resulted: in an increased proliferation rate; increased osteogenic gene expression (RUNX2; p < 0.0001; Osteocalcin; p < 0.0001; and Osterix; p = 0.0037); and faster development of mineralized bone nodules. Discussion/Conclusion : Addition of autologous ALP-PRP in equine BMMSCs and osteogenic cell cultures provided evidence of enhanced bone production in vitro. An increased proliferation rate of BMMSCs can lead to higher numbers of BMMSCs for differentiation into osteoblasts at the fracture site. Increased gene expression can lead to increased production of bone proteins required for bone formation. This study highlights the potential for ALP-PRP to enhance bone repair in horses.
