A disponibilidade de água é um dos principais fatores que influenciam o desenvolvimento e a sobrevivência das plantas.A capacidade das plantas para enfrentar o estresse hídrico é um elemento-chave no seu ciclo de vida e o ácido abscísico (ABA) tem um papel essencial nas respostas a esse e outros estresses abióticos.As quinases SnRK2 (proteínas quinases relacionadas a SNF1) são ativadoras centrais da via de resposta ABA e desencadeiam a resposta ao hormônio por meio de uma cascata de eventos de fosforilação, iniciada com a auto-ativação da quinase.A sinalização intracelular do ABA, bem como a regulação e estrutura das SnRK2s, tem sido amplamente estudada em Arabidopsis thaliana.No entanto, a estrutura e bioquímica dessas proteínas permanecem pouco exploradas em plantas cultivadas, como no caso da cana-de-açúcar.Nesta tese, identificamos as quinases SnRK2 de cana-deaçúcar e estudamos em profundidade sua estrutura, autorregulação e atividade.Surpreendentemente, apesar da alta similaridade entre as estruturas cristalográficas das SnRK2s de cana-de-açúcar e Arabidopsis, nossos estudos de mutagênese indicam que as proteínas apresentam diferenças na sua regulação bioquímica.Além disso, sugerimos um papel diferente para um domínio regulatório da quinase anteriormente relacionado à sua desfosforilação.Além das SnRK2s, focamos no crosstalk entre as vias do ABA e do ácido giberélico (GA), hormônio promotor de crescimento e que apresenta uma interação antagônica com ABA em vários estágios de desenvolvimento da planta.Nesta tese, hipotetizamos que as SnRK2s quinases e as proteínas DELLA -principais repressoras da resposta ao GA -podem ter um papel no conhecido antagonismo entre ABA-GA.Nossas análises, utilizando proteínas de cana e Arabidopsis, demonstraram que SnRK2s e DELLAs são capazes de interagir em leveduras e in planta.Além disso, as SnRK2s de cana foram capazes de fosforilar DELLA in vitro.Essas observações sugerem que SnRK2s e DELLA podem ter um papel no crosstalk entre ABA-GA, possivelmente por meio da fosforilação.
