Demam Berdarah Dengue (DBD) merupakan penyakit berbasis lingkungan (fisik, biologis dan sosisal) yang banyak ditemukan di daerah tropis dan sub tropis yang penularannya melalui gigitan nyamuk spesies Aedes aegypti dan Aedes albopictus. Upaya untuk mencegah penularan penyakit ini dengan memutus rantai perkembangbiakan nyamuk itu sendiri yakni dengan melakukan kegiatan 3M (Menguras, Menutup, Mengubur) plus dan menekan faktor penularan lainnya sehingga sangat berkaitan dengan prilaku. Tujuan penelitian ini adalah untuk mengetahui hubungan kejadian DBD terhadap pengetahuan, kondisi Tempat Penampungan Air (TPA) dan keberadaan jentik nyamuk Aedes sp di wilayah Puskesmas Lingkar Barat Kota Bengkulu. Penelitian ini merupakan penelitian kuantitatif dengan jenis penelitian analitik. Hasil penelitian menunjukkan ketiga variable memiliki hubungan yang bermakna dengan keberadaan jentik Aedes sp, yakni tingkat pengetahuan dengan p value 0,044, kondisi tempat penampungan air dengan p value 0,03 dan keberadaan jentik nyamuk dengan p value 0,035. Sedangkan faktor yang paling besar pengaruhnya/ paling dominan adalah pengetahuan, disusul keberadaan jentik dan tempat penampungan air, dengan nilai Odd Ratio (1,399; 0,623; dan 0,535).
