P142 Symptomatic improvement within 2 days of etrasimod induction therapy for ulcerative colitis in ELEVATE UC 52 and ELEVATE UC 12

Abstract

Introduction Etrasimod is an oral, once-daily (QD), selective sphingosine 1-phosphate (S1P)1,4,5 receptor modulator for the treatment of moderately to severely active ulcerative colitis. This post hoc analysis assessed improvement in rectal bleeding (RB) and stool frequency (SF) in patients (pts) in ELEVATE UC 52 (NCT03945188) and ELEVATE UC 12 (NCT03996369). Methods Daily e-diary data on RB and SF were pooled. Daily Mayo RB and SF subscores (RBS and SFS) and partial modified Mayo score (pMMS; RBS+SFS), and change from baseline (CFB) in the first 28 days of therapy, were calculated. Symptomatic response and remission, and RB and SF remission are reported. Results At baseline, pts receiving etrasimod 2 mg QD and placebo (PBO) had mean (SD) RBS 1.6 (0.69) and 1.6 (0.68), SFS 2.4 (0.74) and 2.4 (0.74), and pMMS 4.0 (1.07) and 4.0 (1.07), respectively. Adjusted differences (etrasimod vs PBO) became significant from Day 2 and 11 for symptomatic response and remission, and Day 15 and 3 for RB and SF remission, respectively (all p<0.05; table 1 Daily responses in pts receiving etrasimod vs PBO on the first day of a significant adjusted difference in response). Conclusions Significant, early improvements indicate a potentially rapid onset of symptomatic effect in pts receiving etrasimod.
