Isolasi Dan Identifikasi Molekuler Bakteri Penghasil Amilase Pada Produk Fermentasi Kentang Yukon Gold (Solanum Tuberosum L.) Berbasis GEN 16S RNA

Abstract
Kebutuhan amilase dan tingginya daya produksi enzim membuat pencarian sumber-sumber amilase yang baru sangat diperlukan salah satunya melalui proses fermentasi Kentang Yukon Gold (Solanum tuberosum, L.) yang mengandung 18,4 g karbohidrat selama 3 hari. Penelitian ini bertujuan untuk mengisolasi bakteri penghasil enzim amilase yang berasal dari produk fermentasi kentang Yukon Gold. Uji penghasilan enzim amilase dilakukan pada media selektif Starch Agar (SA) Uji enzimatik isolat bakteri didapatkan 4 isolat. Proses identifikasi molekuler dilakukan melalui analisis sekuen gen 16S rRNA bakteri yang telah diamplifikasi berdasarkan metode PCR. DNA hasil dari amplifikasi kemudian di sekuensing menggunakan BLAST (Basic Local Aligment Searc Tool), isolate FST-1 merupakan isolat yang memiliki ideks amilolitiki terbesar dengan nilai 2,63. Hasil identifikasi isolate FST-1 didaptkan kemiripan 100% dengan bakteri Pseudomonas sutzeri sehingga isolate FST-1 diberi nama Pseudomonas sutzeri FST-1 (Fermented SolanumTuberosum-1). Kesimpulanya, strain FST-1 merupakan bakteri penghasil amilase yang potensial dan teridentifikasi sebagai Pseudomonas sutzeri srain FST-1.
